How do u know if your blue merle dane has a double or single merle gene??
It really doesn't matter if a Merle (they are only called merle not blue merle) has a double merle gene or not unless you were going to breed, however a merle gene should not be used in a breeding program regardless.

Confused about the Great Dane colors?
I loooong time ago a question "What other races carry merle merle or recessive?" Great Danes I could not carry the merle gene merle danes being dominant because harlequinXharlequin merle crosses produced:. I found this breeder's site "It is, however structurally identical to his brother Spirit and is still as tall as he is y. Annie Baer has been hearing evidence CERF test with both hearing and vision is normal. He has also had a DNA test for the Merle gene. We chose dark horse Harlwood CH (Hoss) and a bull for this ...." When trash says "it has been tested for the gene merle" what really confuses me, what is a harlequin Great Dane. Many people here told me that * NO * dog can "carry" merle, is always dominant. If so, how can two harls produce puppies Merle?

The discovery is most interesting, since it appears that the harlequin gene is not a "color (pigment-producing) gene at all. Simply alters the gene harlequin the effect of the merle gene, so that regions diluted (merle coloration) observed are largely removed from the pattern of the layer, leaving behind the color of the dog every second is known as a harlequin. So now there is proof positive that all the harlequins are genetically merles. The old idea of a "gene switch" remains basically correct and helps the uninitiated understand the interaction of Harl and merle alleles: the "Harl gene acts upon the dogs with the merle mutation to produce a novel phenotype "get the Gray" (ie, elimination of most of all diluted pigments called merle). Harlequins are basically a "Change key" of normal (Mm) merle, in which the Harlequin gene acts in the merle gene to produce the harlequin coat pattern, so Harlequin Great Danes, Neil O'Sullivan eloquent phrase is a "double heterozygotes (HHMM)." Merle mutation is a "dominant" mutation of a gene known as SILV pigment that produces a protein called PMEL17. Merle is a semi-dominant, ie, this gene with two alleles only produces three different phenotypes: not Merle, Merle normal (heterozygous) and the white white (which is homozygous defective commonly referred to as merlikin Danish). The merle gene produces a characteristic increase in white areas and areas of decreased pigment, with patches of pigment tones and full in the heterozygote usually with homozygotes usually mostly white. No merle inherits a copy of the gene Harl. Harl The mutation is also a "dominant" mutation in the wild "type" allele reduces pigmentation. Harl mutation acts as a modifier of merle dogs, further reducing the amount of pigment is seen in the shield. Dogs without a merle gene can lead to gene invisible Harl sight. Both Harlequin and Merle Great Dane has black pigment left behind, because every normal normal Merle and Harlequin have a copy of "wild type" of merle gene from both genes and Harl. A dog with two copies of the mutation Merle and a copy of the mutation Harl appears mostly white with some black and some patches may merle in a pattern known as "white." (Any embyo receiving two copies of the gene seems absorbed Harl.) An important take home message from this research parallels recent knowledge accumulated over many decades breeder that all Harlequin and Merle Great Danes are unique snowflakes, as no two are exactly alike. This is due to the unique nature of the merle gene: for all that all the Harlequins and Merles merle inherit the same mutation, the effect can vary from dog. The coat pattern therefore can not be controlled as to the details of the distribution of pigments (ie, the patterns necessarily vary). How exactly is this effect achieved is currently being explored by the laboratory of Dr. Clark. His previous work includes the discovery of the merle gene, and the isolation of the gene on chromosome 9 harlequin (see PDF and other links below). Charitable trust the DGAC signed the advent of this research through "Harlequin Gene Project" and the Board of Directors recently voted to increase spending on the continued exploration of Dr. Clark harlequin gene and its interaction with the merle gene and others in the Great Dane.
